Anywhere you see a color picker in the UI there will be an alpha channel associated with that.

You can also select the priority NIC in NDI Access Manager but I haven't seen that actually do anything...
Last ditch effort would be to disable the non NDI NIC in device manager, open Arena, Then re-enable that NIC.

You need to setup manual metrics in your OS. If your on Apple I wouldn't know how to do this. For windows:

Yeah, SDI will be the standard for at least 5 more years, most likely 10. From the studio's I've worked in and tech's I've spoken with.
NDI is getting there, close, but not stable enough for a studio IMO. For a small production and tour, sure.

For starters you will need a 25GbE connection PER UHD stream. So laptops are now out of the question. Most touring media servers have 2.5Gbe connections at best 10Gbe is you're super lucky.
