My apologies, I should have included that I am able to get 2 universes out of Resolume by creating 2 Lumiverses in the advanced output and creating 2 DMX outputs in the preferences window, just not with this Enttec device.
I have output 2 universes by using the 1 output available from the Enttec Mk2 and 1 output over Artnet using a different Enttec device (ODE). When I create more than 1 output of DMX in the preferences tab, only port 1 of the Mk2 shows as an option for any of the outputs. Picked up the Mk2 to output 2 universes and not carry the ODE.

You're currently stuck in development limbo between us, Enttec and Apple.
Here's the story. Please bear with me.
Long ago, we used Apple's own USB drivers to connect to Enttec devices. This was confusing to a lot of users, because Enttec themselves said to use a different driver set. So we changed it. We were in sync. Things were good. It was a happy time.
Then, around the release of Mavericks. Apple decided to only allow their own set of drivers, and disable all apps that use different drivers. This included Enttec. Enttec didn't know what to do for the longest time. Even their own website still says the USB Pro MK1 and MK2 will not work on El Capitan. But, when you download Pro-Manager, it says their devices are now working with Apple's own drivers on OSX.
For us, this means we have to go back to our old code. This is going to take a little while.
In the meantime, I suggest to contact Enttec and ask them how to use their devices on OSX with software that uses their old driver set. If they get back to you, please let us know as well. We have a very hard time getting support from them.

The good news is that on El Capitan, Enttec's suggestion to use Terminal to enable the old drivers seems to work for me. I can see the old Mk1 and both ports on the Mk2 after running it.
sudo kextunload -b com.apple.driver.AppleUSBFTDI
Edit 27/6/2016: Don't do this anymore! Just update to El Capitan and Arena 5.0.2 and you'll be fine. Sudo'ing your way around things will probably break your computer.

@Janis I am having the problem of the 2nd port not being an option in DMX output of Arena 5.. it just doesnt recognise that there's another port for the PRO MK2.. can only run one universe. Have installed all the latest drivers, firmware.. tried disabling drivers via terminal etc.. All Ok in the pro manager software. Have even spoken with Enttec tech support & they recommended contacting you guys. Running El Capitan & V5.0.4.. Any ideas?

Sorry to bring up an older post, Just curious if the Enttec USB to Artnet boxes are supported in the Windows version of Resolume? Have a client with a small cheap console that does not accept Artnet and wanted to find a DMX solution for them.

I don't think there's such a thing as an USB to Art-Net box.
Just use literally any Art-Net to DMX node. You'll send Art-Net out of Resolume to the node, and then send DMX from the node to the desk.

The client was looking for a cheap solution to send DMX from Resolume without the use of Artnet.

Yeah, my point is that even when the desk doesn't support Art-Net, you can still use Art-Net out of Resolume and convert that to DMX to send to the desk.
You're going to have to convert using some box at one point, so it's either
Resolume > USB > converter > DMX > desk
or
Resolume > Art-Net > converter > DMX > desk
Why go with some propietary USB protocol if you can use an industry standard?
